Micropropagation Technology through Tissue Culture
Introduction
Standardized protocols are available to distinguish hybrids from the of cotton, sunflower, castor, rice and sorghum using protein or isozyme and DNA markers.
Application
These techniques are applicable for testing genetic purity of hybrid seeds. The purity of CMS female parent can also be tested by RAPD analysis.
Process
Isoenzymes are extracted from 100 single seeds or seedling and analyzed by electrophoresis. DNA is extracted from 100 single seedlings and analyzed by RAPD using specific primers.
